表观遗传学是指基于非基因序列改变所致基因表达水平变化。表观遗传的现象目前已知的包括DNA甲基化、基因组印记、母体效应、基因沉默、核仁显性、休眠转座子激活和RNA编辑等。
其中组蛋白(Histone)的修饰作用是表观遗传学研究的一个重要领域。Histone H3与H2A、H2B和H4一起参与真核细胞中染色质的结构。组蛋白H3通过承受几种不同类型的表观遗传修饰,参与并影响许多细胞进程,如转录激活/失活、染色体包装、DNA损伤/修复。这些修饰,包括甲基化、瓜氨化、乙酰基化、磷酸化、小泛素相关修饰化、泛素化及二磷酸腺苷核糖基化。主要发生在组蛋白H3的N末端尾区域上,这导致核小体结构重构成更容易接近的开放构象的转录复合物。在大多数物种中,组蛋白H3主要在赖氨酸9,14,18,23和56处乙酰化; 在赖氨酸4,9,27,36和79处甲基化; 并分别在ser10,ser28,Thr3和Thr11磷酸化。组蛋白修饰的深入研究能更好地了解细胞过程的表观遗传调控。
